CHO PRDX1 ELISA
PRDX1 (Peroxiredoxin-1) is recognized as a high-risk Host Cell Protein (HCP) due to its strong immunogenic potential, redox activity, and notorious “hitch-hiker” behavior. As a thiol-dependent peroxidase that regulates intracellular ROS, PRDX1 plays roles in signaling pathways (e.g., NF-κB, MAPK) and stress responses. When present as a process-related impurity, these properties make PRDX1 a concern for product quality and patient safety.
Its industry relevance is supported by multiple studies. PRDX1 was detected across a broad set of commercial biologics (Molden et al., 2021; Falkenberg et al., 2018) and several reports highlight immunogenicity risks associated with PRDX1 and related redox-active proteins (Albrecht et al., 2018; Jawa et al., 2016; Joudal et al., 2013; Park et al., 2017). In process development, Guo et al. (2023) identified elevated PRDX1 by LC-MS/MS, noted measurement challenges with some other commercial kits (dilution non-linearity/spike-recovery in one matrix), and showed process optimization could reduce PRDX1 to <5 ppm by MS-guided control strategies.
Together, these findings have led regulators to encourage risk-based HCP control with protein-specific monitoring for high-risk impurities like PRDX1.
BioGenes PRDX1 ELISA Approach
BioGenes now offers a CHO-specific ELISA kit purpose-built for targeted monitoring of PRDX1 across process intermediates and drug substance. The kit uses recombinant CHO PRDX1 expressed in HEK293. This approach minimizes residual CHO contaminants during antigen preparation and reduces undesired cross-reactivity to other CHO HCPs. The assay was developed with high-quality polyclonal antibodies, providing broad epitope coverage and reliable recognition of native PRDX1. The assay is compatible with both in-process samples (cell culture supernatants, intermediates) and drug substance.
