Monoclonal Antibodies

Q:

Are polyclonal antibodies or monoclonal antibodies more suitable?

A:

Please see the following overview:

Advantages

Disadvantages

Polyclonal antibodies

inexpensive

short preparation times (normally 6-8 weeks)

limited availability

mixture of antibodies having differing properties (constants, epitope binding)

cannot be reproduced or can only be reproduced with difficulty

Monoclonal antibodies

always reproducible and can be prepared in any desired quantity

possibility of selecting antibodies which possess very specific properties

cost-intensive preparation

processing time of up to 6 months

Q:

Which animals are used?

A:
  • Balb-C mice
  • Brown Norway rats

Other mice or rats on request.

Q:

Which kind of antigens can be used?

A:
  • proteins
  • peptides
  • peptide conjugates
  • haptens
  • hapten conjugates
  • small molecules
  • toxic molecules
Q:

In what form can the antigen be supplied?

A:
  • lyophilised
  • in buffer (concentration 0,5 -1 mg/ml)
Q:

What should be the purity of the antigens?

A:
  • proteins: > 95%
  • peptides: > 95%
Q:

What quantities of antigen are required?

A:
  • protein, at least 1 mg
  • peptide, at least 5 mg

The prospects for obtaining a good immune response increase in proportion to the amount of antigen available. If other quantities or concentrations are available, please contact us. We shall find a solution.

Q:

Which antigen contamination are not suitable?

A:

SDS, urea, guanidine and other toxic substances

Q:

What does the guarantee offer include?

A:

The guarantee offer covers the following services:

  • at least three positive stable clones producing IgG aginst the used immunogen
  • positive reaction of IgG refers to the test system used by BioGenes (ELISA)
  • customer pays only in case of success
Q:

When can advantage be taken of the Guarantee Offer?

A:

At least 1.5 mg of soluble antigen (95% purity) or 4-5 mg of peptide which is present at a concentration of 1mg/ml are required.

Antigens which are in a gel or in suspension, or only available in a quantity of less than 1.5 mg, and also peptide conjugates supplied by the customer, are not suitable.

Q:

Are other offers possible?

A:

Depending on the nature and quantity of the antigen, and on the wishes of the customer, it is possible to put together package offers which do not contain any guarantee but which are only appropriately invoiced in accordance with the work involved.

Q:

What is supplied to the customer?

A:

Quick and Easy Offer

  • 10 ml cell culture supernatant and 3 cryo cultures from each stable clone (up to 5) after cloning

Modular Offer

  • 1 ml cell culture supernatant from each positive clone after 1st cloning for testing purposes (up to 10)
  • 10 ml cell culture supernatant and 3 cryo cultures from each stable clone (up to 10) after 2nd cloning

Guarantee Offer

  • 1 ml cell culture supernatant from each positive clone after 1st cloning for testing purposes (at least 3, up to 10)
  • 10 ml cell culture supernatant and 3 cryo cultures from each stable clone (at least 3, up to 10) after 2nd cloning
Q:

Who owns the clones?

A:

The client is always the owner; BioGenes will not use the clones in any other way unless there is an agreement with the client in this regard.

Q:

Can cryocultures be stored with BioGenes?

A:

Storage in liquid nitrogen, and maintenance of the clones, is possible by arrangement.

Q:

Is it possible to produce larger quantities of antibodies?

A:

We can prepare quantities of up to the gram scale both from the clones which we have developed ourselves and from clones which are supplied to us (must be accompanied by a certificate that the clone was tested negative on mycoplasm).
